Protein L is an immunoglobulin-binding protein originally derived from the surface of bacterium Peptostreptococcus magnus. Protein L binds to Ig through L chain interaction from which the name was derived. The native Protein L consists of 719 amino acid residues with the molecular weight estimated as 95 kDa by SDS-PAGE analysis. Protein L does not contain any inter chain disulfide loops, nor does it consist of disulfide-linked subunits. This version of Protein L is now produced recombinantly in our unique MPTxpress liquids-free, fully single-use E.coli manunfacturing platform.

Recombinant Protein L is expressed by MicroProtein Technologies in E.coli using its MPTxpress upstream platform with innovative purification methods. MicroProtein’s Recombinant Protein L is a non-glycosylated polypeptide having a molecular mass of 41 kDa and comprised of 5 IgG-binding regions of Protein L (B1-B5). The cell wall-binding region, cell membrane-binding region and albumin-binding regions have been eliminated from this version of recombinant Protein L to ensure maximum specific IgG binding. Protein L has the unique ability to bind through kappa light chain interaction without interfering with an antibody antigen-binding site. This gives Protein L the ability to bind a wider range of Ig classes compared to Proteins A, G, etc. Protein L will bind to all classes of Ig (IgA, IgD, IgE. IgG and IgM). Protein L will also bind single chain variable fragment (scFV), Fab, and Dab fragments. Protein L binds kappa 1, 3, 4 in general, making it optimal for purification of many newer engineering antibodies.
Recombinant Protein L is valuable as the basis for preparing various kinds of affinity media or probes for detection or purification of human and mouse antibodies in immunoassays and antibody purification protocols.
